Selank (TKPRPGP) is a synthetic heptapeptide analogue of the endogenous immunomodulatory tuftsin (Thr-Lys-Pro-Arg) with a Pro-Gly-Pro C-terminal extension; preclinical and clinical studies demonstrate anxiolytic activity comparable to benzodiazepines but without hypnosedative effects, mediated in part through positive allosteric modulation of GABA-A receptors in a subtype-selective, concentration-dependent manner (https://pubmed.ncbi.nlm.nih.gov/30255741/). GABAergic mechanism studies show Selank affects [3H]GABA binding and also modulates dopaminergic and serotonergic systems, while its effects on the sleep-wakefulness balance (upregulating Hcrt expression) explain the absence of sedation seen with classical benzodiazepines (https://pmc.ncbi.nlm.nih.gov/articles/PMC4757669/).
