Research Mechanism Summary
PTD-DBM is a fusion peptide consisting of a protein transduction domain (PTD) for membrane penetration and a Dishevelled (Dvl) binding motif (DBM) for intracellular targeting. Once internalized, DBM competitively blocks the interaction between CXXC5—a negative feedback regulator of Wnt signaling—and Dvl, thereby disinhibiting the Wnt/β-catenin pathway. In murine androgenetic alopecia models, PTD-DBM promoted new hair follicle formation and stimulated the anagen phase through upregulation of β-catenin, alkaline phosphatase, and PCNA (https://biotechpeptides.com/product/ptd-dbm-5mg/). Co-administration with valproic acid (a GSK3β inhibitor) produced synergistic hair regrowth and new follicle neogenesis in vivo.
