Adipotide (FTPP) is a chimeric peptidomimetic (CKGGRAKDC-GG-D(KLAKLAK)₂, MW ~2557 Da) engineered by combining a targeting motif (CKGGRAKDC) with a proapoptotic domain (D(KLAKLAK)₂) via a Gly-Gly linker. The targeting motif binds a membrane receptor complex involving annexin A2 (ANXA2) and prohibitin (PHB) enriched on endothelial cells in white adipose tissue vasculature; following receptor-mediated internalization, the amphipathic (KLAKLAK)₂ domain disrupts mitochondrial membranes, triggering apoptosis in fat-associated endothelial cells. In obese rhesus monkey studies, Adipotide administration produced measurable reductions in white adipose tissue and improved insulin sensitivity, establishing a mechanistic proof-of-concept for vascular-targeted proapoptotic peptides as research tools for studying adipose tissue biology.
