GHRP-6 is a hexapeptide (His-D-Trp-Ala-Trp-D-Phe-Lys-NH2) that was among the first synthetic peptides characterized as a potent GHS-R1a agonist; it stimulates pituitary GH secretion and also acts at hypothalamic arcuate nucleus neurons to amplify endogenous GHRH-driven GH pulses (https://pubmed.ncbi.nlm.nih.gov/11322495/). Research demonstrates its activity on gastric ghrelin-producing cells as a partial mechanism of systemic GH effects, and ghrelin — the endogenous GHS-R1a ligand — was later identified in part through work with GHRP-6 pharmacology (https://pubmed.ncbi.nlm.nih.gov/11322495/).
